The Decomposition of Yeast Nucleic Acid by a Heat Resistant Enzyme.
نویسنده
چکیده
The yeast nucleic acid used in these experiments was a commercial preparation purified by treatment with picric acid (3). A sample of yeast nucleic acid and one of thymus nucleic acid were kindly supplied by Dr. P. A. Levene to whom the author wishes to extend his heartiest thanks. The effect of the enzyme preparations on nucleic acid was followed by determining the amount of total phosphorus rendered soluble in hydrochloric acid during the incubation of the enzymesubstrate mixture. To this end, the mixture was treated with HCl (final conoentration 0.2 ~)j and then immediately cooled and centrifuged in the cold. The supernatant was then analyzed for phosphorus by the Youngburg and Youngburg method (4) modified in details. It was found necessary to prolong the digestion with sulfuric acid for 40 minutes in order to obtain a complete liberation of the phosphorus in the inorganic form. MacFadyen followed the decomposition of nucleic acid by deter-

around pH 4 to pH 5; even at pH 1.5 and pH 7.5 the enzyme can be heated in a boiling water bath for five minutes with only little loss of activity. The rate of action upon yeast nucleic acid increases with temperature up to 750 C.; it then decreases sharply and no appreciable action can be detected at 850 C. When an enzyme-substrate mixture is maintained at 950 C. no enzymatic hydrolysis occurs...
متن کاملCellular Morphology and Immunologic Properties of Escherichia coli Treated With Antimicrobial Antisense Peptide Nucleic Acid
Background & Objectives: Antisense peptide nucleic acids (PNA) that target growth essential genes show potent bactericidal properties without cell lysis. We considered the possibility that whether PNA treatment influence the bacteria total nucleic acids content and apply approach to develop a new delivery system to Dendritic cells (DCs). DCs are the most potent antigen presenting cells in th...

Development of potent new anti-Shigella agents for rapid and specific detection and treatment is of great importance. Aptamers, nucleic acid oligomers capable of specific binding to a wide range of non-nucleic acid targets, may be of value for this purpose. In the present study, we used a Systematic Evolution of Ligands by Exponential enrichment (SELEX) process to select DNA aptamers that b...
متن کاملCrystalline Ribonuclease
1. A crystalline enzyme capable of digesting yeast nucleic acid has been isolated from fresh beef pancreas. 2. The enzyme called "ribonuclease" is a soluble protein of albumin type. Its molecular weight is about 15,000. Its isoelectric point is in the region of pH 8.0. 3. Ribonuclease splits yeast nucleic acid into fragments small enough to diffuse readily through collodion or cellophane membra...
متن کاملReduction of nucleic acid content in Candida yeast cells by bovine pancreatic ribonuclease A treatment.
Yeast as a source of protein for human consumption is limited by its relatively high nucleic acid content. In this study, we developed an enzymatic method of decreasing the nucleic acid content. Candida utilis cells, heat-shocked at 80 C for 30 sec, were treated with bovine pancreatic ribonuclease A. Maximum leakage of nucleic acid was observed when the incubation temperature was between 55 and...
